This article explores the mechanism of action, clinical applications, challenges and future prospects of immune checkpoint inhibitors in cancer therapy. Despite initial responses to ICIs, many patients develop resistance over time, leading to disease progression. Resistance mechanisms may involve adaptive immune evasion, tumor cell intrinsic factors and alterations in the tumor microenvironment. Understanding these mechanisms is crucial for developing strategies to overcome resistance and prolong the durability of response.
