Advances In DNA Sequencing

DNA sequencing is the process of verifying the precise order of nucleotides inside a DNA molecule. It is used to determine the order of four bases adenine , guanine , cytosine and thymine in a strand of DNA.

BASIC METHODS:

•             Sanger sequencing method

•             Maxam and Gilbert sequencing method

These represents the first GENERATION of DNA sequencing methods.

In the late 20 and early 21st century, efforts have been made towards the development of new methods to replace the first generation technology.

The new methods are referred to as next generation sequencing and their use has changed the scientific approaches in both basic and applied research .

Next generation DNA Sequencing

•             It is a new method for sequencing genomes at high speed and at low cost.

•             It is also known as second generation sequencing or massively parallel sequencing.

It is also used to describe number of different modern sequencing technologies including

•             Illumina sequencing

•             Pyrosequencing

•             Ion torrent sequencing

•             SOLID sequencing

The difference is that instead of sequencing a single DNA fragment, New generation sequence extends this process across millions of fragments in massively parallel sequencing.

More than 90% of world’s sequencing data are generated by Illumina sequence
