Food can alter the absorption, distribution, metabolism and excretion of drugs, leading to variability in therapeutic efficacy and safety. This article reviews strategies to overcome food effects on drug bioavailability from both formulation and biopharmaceutical perspectives. Emphasis is placed on the formulation approaches such as food-modulated drug delivery systems and the biopharmaceutical strategies involving the manipulation of drug characteristics to mitigate food effects.

Food intake can significantly influence drug absorption and bioavailability, often resulting in unpredictable pharmacokinetics and variability in drug efficacy and safety. The complexity of food effects arises from various factors including changes in gastric pH, gastric emptying rate, bile secretion and enzyme activity.
