Black pepper (Piper nigrum L.) known as the ?king of spices? is the World?s most important commercial spice which originated in India. Now it is widely cultivated across various parts of the world. Unambiguous characterization of cultivars and selected germplasm is an urgent requirement in tune with the globalization of agriculture. Of the several PCR techniques developed during the last two decades, random amplified polymorphic DNA (RAPD) and inter simple sequence repeats (ISSR) offers a simple and economical means for genotype characterization. The information obtained through molecular marker characterization is extensively used for identification of genotypes, screening duplicates, assessing genetic diversity and monitoring the genetic stability of conserved germplasm. The present study deals with DNA fingerprinting of Piper nigrum cultivars using molecular markers. Seven released varieties of Piper nigrum (Panniyur1- Panniyur7) were used for the analysis. DNA isolation technique was standardized using CTAB method. Good quality DNA with UV absorbance ratio (A260/A280) 1.80 - 1.95 was used for analysis. Forty decamer primers and fifty ISSR primers from Operon Tech were screened for the purpose. The varieties were further characterized using the 10 selected primers. Fingerprints were developed for all the seven varieties and were documented. The data generated and the fingerprints developed would be of great use for testing the clonal fidelity.
