Effect of antioxidants on biostabilization of RBC

Abstract :

One of the least recognized causes of cellular damage during ex vivo preservation of red blood cells is oxidative injury to the hemoglobin. Hemoglobin oxidation largely dictates our ability to effectively preserve red blood cells. Understanding its origins along with investigating methods to minimize it can significantly improve the quality of our future blood products. Oxidative injury in stored RBCs is more likely to be the result of crosslinks between denatured Hb and membrane components. Vitamin E and C are believed to function as an antioxidant in vivo protecting membranes from lipid peroxidation. In several studies it has been reported that the exogenous supplementation of vit E, C and B3 reduced the oxidative damage of the stored RBCs . The purpose of the present study is 1) to investigate the protective effect of the various antioxidants like α- tocopheral, ascorbic acid, nicotinic acid, retinol and phylloquinone against the oxidative damage of RBCs during freeze drying and freeze thawing and 2) to compare the effect of these antioxidants. For doing this, various cellular integrity tests like the cell viability test, hemoglobin oxidation assay and oxidative damage assays were performed at each stage of cell processing. The results obtained by us showed the reduced levels of metHb in the presence of antioxidants indicating their protective effect.
