Abstract :

Novel cyclic analogues of enkephalin containing xylene bridge regioisomers (7a-c) have been synthesized and fully characterized as mixed �?¼, opioid receptors agonists.a,b The in vitro activity has been investigated showing a good affinity of 7a-c for both of them. In vivo biological assays revealed that 7b is the most potent cyclic analogue with the ability to maintain high level of analgesia after 60 and 90 minutes following i.c.v. and i.t. administrations respectively in Tail Flick test and long lasting analgesia after subcutaneous administration.
